how to calculate theoretical net peptide content?

Most of the impurities in the polypeptide sample are counterions, and a small part contains water, solvents absorbed by the polypeptide, and some other substances. Assuming that the counterion trifluoroacetate (TFA) is the only non-polypeptide impurity in the polypeptide product delivered to you, the theoretical net polypeptide content can be estimated:

first, we divide the molecular weight of the polypeptide into two parts, one is the polypeptide and the other is the TFA counter ion of the neutralizing polypeptide. The molecular weight of TFA can be calculated by multiplying the number of ions by the molecular weight of TFA counter ion (MW = 114). If the molecular weight of the synthetic polypeptide is 1000, and the polypeptide has a free N-terminus and an Arg residue, the theoretical net polypeptide content is calculated as 1000/(1000+2 x 114 ) = 1000/1228=0.81 or 81%. Therefore, our actual polypeptide net content can usually be obtained by quantitative amino acid analysis.
